چکیده

Isolated mammalian metaphase chromosomes suspended in ion-free or 10d3 YN&l solution undergo an irreversible, pH-dependent structural change, as reflected by electrometric titrations, pH-stat experiments and measurements of circular dichroism spectra. Unmasking of titratable groups with pK’s lower than the isoionic pH of chromosomes in ion-free water, pH 66, accompanies the change in conformation. The structural change is inhibited in aqueous solutions of O-001 Mmagnesium or calcium ion. Adding magnesium or calcium ions to a final concentration of 10m3 Y results in a lowering of the pH of the water-washed chromosome solution from 56 to 4.9. The binding constant for magnesium ion to chromosomal sites was measured to be 1-l x 106 (moles/liter)-r and the number of binding sites is 16 x lows millimole per milligram of chromosomes.

In vitro study of drug-protein interaction using electronic absorption, fluorescence, and circular dichroism spectroscopy

In the near future, design of a new generation of drugs targeting proteins will be required. Considering the complex bond between the drug and protein, the structure and stability of the target protein should be considered. So far, a series of in vitro investigations have been conducted with the aim of predicting drug-biological medium interactions. In these studies, use of spectroscopic method...

Investigation of spectral conversion of d(TTAGGG)4 and d(TTAGGG)13 upon potassium titration by a G-quadruplex recognizer BMVC molecule

We have introduced a G-quadruplex-binding ligand, 3,6-bis(1-methyl-4-vinylpyridinium)carbazole diiodide (BMVC), to verify the major structure of d(T2AG3)4 (H24) in potassium solution and examine the structural conversion of H24 in sodium solution upon potassium titration. The studies of circular dichroism, induced circular dichroism, spectral titration and gel competition have allowed us to det...

Magnesium-binding studies reveal fundamental differences between closely related RNA triphosphatases

The Chlorella virus RNA triphosphatase (cvRTPase) is involved in the formation of the RNA cap structure found at the 5'-end of the viral mRNAs and requires magnesium ions to mediate its catalytic activity. To extend our studies on the role of metal ions in phosphohydrolysis, we have used a combination of fluorescence spectroscopy, circular dichroism, denaturation studies and thermodynamic analy...
